auto-heal
multiplayer (they might do it somehow, you never know)
I really hope they have avoided those things. There's too much focus on multiplayer these days and auto-heal is lame.

Multiplayer wouldn't be too creative. Assassin's Creed's multiplayer was merely a copy of other multiplayers with a rating system similar to Hitman's.
Auto-Heal wouldn't hurt. As it has been said. Maybe in easier difficulties but not in harder ones.
Overall, by asking them not to change the mechanics, is asking for horrible reviews.
A cover system could be employed if done right. Complete with optional rating system.
I don't want this game to become more like Assassin's Creed. With the Automatic Desynchronization or Death if you are caught.
Players in Blood Money found themselves shooting their ways through the rest of the mission if they were caught. Starting the level over always fixed the rating and will in future games regardless of the cover system or auto-heal.

Only multiplayer i would accept is a co-op campaign. Maybe 47, and another hired assassin to help you.
